Institut Curie is a major player in the research and fight against cancer. It consists of a Hospital group and a Research Center of more than 1000 employees with a strong international representativeness.

The objective of the Research Center is to develop basic research and to use the knowledge produced to improve the diagnosis, prognosis, and therapeutics of cancers as part of the continuum between basic research and innovation serving the patient.

Laboratory

The Pouponnot’s team investigates signaling pathways and their impact on transcription factors in cancer, employing a range of biochemical and functional experimental approaches both in vitro and in vivo. We have extensive expertise in the use of mouse models and have developed a strong research focus on pediatric brain tumors, particularly medulloblastoma, as demonstrated in our publications (Garancher et al., Cancer Cell, 2018; Morabito et al., EMBO Mol Med, 2019). Our group has established genetically engineered mouse models, patient-derived xenografts, and advanced radiobiology tools to study cancer, including pediatric brain tumors, especially medulloblastoma (MB). Current research efforts are focused on elucidating mechanisms of treatment resistance—especially to radiotherapy—and identifying actionable molecular vulnerabilities in pediatric brain cancers.

The project

Medulloblastoma (MB) is a pediatric tumor of the cerebellum, which represents the most frequent malignant brain tumor in childhood. It is classified into four subgroups: WNT, SHH, group 3 (G3) and group 4. G3 has the poorest prognosis and is associated with a high rate of metastasis and a high probability of relapse. Mechanisms and transcription factors (TFs) that drive its initiation, maintenance, therapeutic resistance and metastatic potential are poorly characterized. Here, we propose to investigate the role of a TF recently identified as a major regulator of the G3 signature. The project focuses on the understanding of its role in G3-MB at the molecular level (RNAseq, ChIPSeq, CRISPR Screen (TF network)…), cellular level (proliferation, protection against apoptosis, migration…) and in tumor progression (in vivo orthotopic grafting).

-        work outside regular working days (in vivo experiments)

Training and Skills required

-        Training: PhD with strong experience in molecular and cellular biology as well as in vivo experiments. Knowledge in cancer biolo  

-        Scientific skills: Cell culture (cell lines and primary cells)/ In vivo mouse experiment/ molecular biology (Cloning, CRISPR, RNA DNA preparation…) / Biochemistry (WB, IP, ChIPSeq…)

-        Professionnal experience desirable: PhD

-        Language skills: ability to write and communicate in English

Desirable Knowledge, skills and abilities:

-         Preferably with experiences in 3D cell culture and primary cell culture

-         In vivo experimentation

-         Competence in basic NGS data analyses and statistical methods is a strong merit but not required.

Contract information

Type of contract: Fixed-term contract.

Starting date:  1st October 2025

Duration: 3 years

Working time: full time

Remuneration: according to the current grids of the Institut Curie and to the experience of the candidate
